A ordem dos Passeriformes é uma das mais pressionadas pelas ações antrópicas, especialmente as relativas ao tráfico de animais, que, devido às más condições de manejo e higiênico-sanitárias, favorecem a infecção dos espécimes por patógenos virulentos e zoonóticos, como cepas de Escherichia coli e Salmonella spp., cujo isolamento em suabes cloacais, bem como a análise dos genes de virulência das cepas de E. coli foram objetivos do estudo. Para isso, 120 Passeriformes silvestres nativos, recebidos pelo Cetas/CE, foram avaliados individualmente. As cepas isoladas foram submetidas a teste de disco difusão para determinação da sensibilidade aos antimicrobianos. Em etapa posterior, foi realizada PCR para a detecção de oito genes de virulência dos principais patotipos diarreiogênicos de E. coli. Quanto aos resultados, nenhuma cepa de Salmonella spp. foi isolada, no entanto a ocorrência de E. coli foi de 40,8%. Foi observada elevada resistência, principalmente aos antimicrobianos tetraciclina, ampicilina e sulfazotrim, ocorrendo multirresistência em 42,8% das cepas. Pela análise molecular, foram diagnosticados quatro entre os nove genes pesquisados, com a identificação de EPEC típicas, EPEC atípicas, ETEC, EHEC e EAEC. Os resultados apontam para a importância de Passeriformes como possíveis disseminadores de zoonoses.(AU)


The order Passeriformes is one of the most pressured by anthropic actions, especially those related to animal trafficking. Due to poor sanitary and hygienic conditions, the infection of the specimens is favored by virulent and zoonotic pathogens such as strains of Escherichia coli and Salmonella spp., whose isolation in cloacal swabs as well as the analysis of the virulence genes of E. coli strains were the objectives of the study. For this, 120 native wild Passeriformes, received by CETAS/CE were individually evaluated. The isolated strains were submitted to diffusion disc test to determine sensitivity to antimicrobials. In a later stage, PCR was performed for the detection of eight virulence genes from the main E. coli diarrhoeagenic pathogens. Regarding the results, no strain of Salmonella spp. was isolated; however, the occurrence of E. coli was 40.8%. High resistance was observed, mainly to the antimicrobials Tetracycline, Ampicillin and Sulfazotrim, with multi-resistance in 42.8% of the strains. By molecular analysis, four of the nine genes were diagnosed, identifying typical EPEC, atypical EPEC, ETEC, EHEC and EAEC. The results point to the importance of Passeriformes as possible disseminators of zoonoses.(AU)

This study aimed to identify serogroups of Escherichia coli important for human health in isolates from psittacine of illegal wildlife trade in Ceará State. In addition, hemolysis and production of Extended Spectrum Beta-Lactamases (ESBL) was assessed in the isolates. A total of 78 E. coli strains isolated from different Psittaciformes species from a wildlife rehabilitation center in Fortaleza, Brazil. The isolates used in this study were previously identified and stored. Serogroup identification was performed using polyvalent sera for EPEC (O55, O111, O119, O114, O125, O86, O126, O127, O128), EIEC (O136, O124) and EHEC (O157). ESBL detection was performed with double disk synergy method. For hemolysis detection, isolates were inoculated in blood agar base enriched with ovine blood. Only 31 (39.7%) isolates were seropositive and the most frequent were O127, O114, O128 and O111. There was no agglutination for serogroups O55, O124, O136 or O157. Considering both seropositive and seronegative isolates, 9 (11.5%) and 35 (44.9%) presented hemolysis and ESBL production, respectively. In conclusion, the investigated psittacine from illegal wildlife trade hosted ESBL-producing E. coli strains and some belong to important serogroups often linked to severe human infections.(AU)


Este trabalho teve como objetivo identificar sorogrupos de E. coli importantes para a saúde humana, oriundos de psitacídeos provenientes do tráfico no estado do Ceará, assim como detectar atividade hemolítica e produção de betalactamase de espectro estendido (ESBL). Foram testadas 78 cepas de Escherichia coli provenientes de psitaciformes do Centro de Triagem de Animais Silvestres, Fortaleza, CE. Para a identificação dos sorogrupos, utilizaram-se soros polivalentes EPEC (O55, O111, O119, O114, O125, O86, O126, O127, O128), EIEC (O136, O124) e EHEC (O157). Para detecção de ESBL, as cepas foram submetidas ao método de aproximação de disco e, para a detecção de hemolisina, foram plaqueadas em ágar sangue base enriquecido com sangue de carneiro. No geral, 31 (39,7%) das amostras foram soropositivas. Os sorogrupos mais frequentemente detectados foram O127, O114, O128 e O111. Não houve positividade para os sorogrupos O55, O124, O136 e O157. Considerando-se as amostras sororreagentes e não sororreagentes, observou-se que nove (11,5%) e 35 (44,9%) cepas de E. coli apresentaram produção de hemolisinas e de ESBL, respectivamente. Em conclusão, constatou-se que psitacídeos provenientes do tráfico de animais silvestres albergam cepas de E. coli produtoras de ESBL e providas de importantes sorogrupos implicados em graves infecções humanas.(AU)

This study was conducted to detect enterobacteria in captive Grey-breasted parakeet from a commercial breeding located in the state of Ceara. 25 individual cloacal swabs were collected and this material was subjected to microbiological processing to obtain the bacterial isolation, identification, and evaluation of the sensitivity profile to antimicrobials. The bacterial isolation in ten samples was determined, belonging to the genera Enterobacter, Hafnia, Serratia, Escherichia, Pantoea and Klebsiella, in which these last three corresponded to the bacteria most isolated, with respective percentages of 28.6%, 21.4%, and 21.4%. As for the sensitivity profile, antibiotics to which the bacteria showed more resistance were trimethoprim-sulfamethoxazole (57%), tetracycline (21%), and azithromycin (43%). Thus, it was shown that the parakeets were carriers of Enterobacteriaceae with resistance characteristics.(AU)

In the development of new materials for orthopedic implants, special attention has been given to Ti alloys that show biocompatible alloy elements and that are capable of reducing the elastic modulus. Accordingly, Ti-Nb-Si alloys show great potential for application. Thus, this is a study on the microstructures and properties of Ti-35Nb-xSi alloys (x=0, 0.15, 0.35 and 0.55) (wt%) which were thermally treated and cooled under the following conditions: furnace cooling (FC), air cooling (AC), and water quenching (WQ). The results showed that Si addition is effective to reduce the density of omega precipitates making beta more stable, and to produce grain refinement. Silicides, referred as (Ti,Nb)3Si, were formed for alloys containing 0.55% Si, and its formation presumably occurred during the heating at 1000°C. In all cooling conditions, the hardness values increased with the increasing of Si content, as a result from the strong Si solid solution strengthening effect, while the elastic modulus underwent a continuous reduction due to the reduction of omega precipitates in beta matrix. Lower elastic moduli were observed in water-quenched alloys, which concentration of 0.15% Si was more effective in their reduction, with value around 65 GPa. Regarding Ti-35Nb-xSi alloys (x=0, 0.15 and 0.35), the "double yield point" phenomenon, which is typical of alloys with shape memory effect, was observed. The increase in Si concentration also produced an increase from 382 MPa to 540 MPa in the alloys' mechanical strength. Ti-35Nb-0.55Si alloy, however, showed brittle mechanical behavior which was related to the presence of silicides at the grain boundary.

RESUMO

The present study reports the isolation of Salmonella enterica in organs of free-living domestic pigeons. In the clinic examination, the presence of feces in the peri-cloacal and abdominal regions were observed, as well as symptoms such as cachexy, incoordination and opisthotonos. Before any therapeutic protocol was applied the bird died and a necropsy was then performed for the removal of spleen, liver, kidney and intestine for bacteriological examination and antibiotic sensitivity test. Salmonella enterica subsp.enterica (O:4,5:i-) and Salmonella enterica subsp. enterica serovar Typhimurium were isolated from the liver and intestine and the sensitivity test demonstrated that these strains are sensitive to several antibiotics...


O presente trabalho relata o isolamento de Salmonella enterica em órgãos de um pombo doméstico de vida livre. No exame clínico foi observada a presença de fezes pericloacal na região ventral, caquexia, incoordenação motora e opistótono. Antes de iniciar um protocolo terapêutico, a ave foi a óbito, e, em seguida, foi realizada uma necropsia para remoção do baço, fígado, rim e intestino para exame bacteriológico e teste de sensibilidade a antibióticos. Foi isolado Salmonella enterica subsp. enterica (O:4,5:i-) e Salmonella enterica subsp. enterica sorovar Typhimurium do fígado e intestino, e o teste de sensibilidade demonstrou que essas cepas são sensíveis a vários antimicrobianos...

This study showed a low prevalence of Salmonella spp. in captive psittacines from zoos and a commercial establishment of Fortaleza. None of the isolated serotypes (S. Lexington, S. Saintpaul and S. Newport) have yet been reported in Amazona aestiva, Ara chloroptera or Melopsittacus undulatus. However, the fact that most birds presented negative for Salmonella spp. may not imply the absence of this pathogen in these birds, since the intermittent excretion is a well-known characteristic of this microorganism...


A manutenção de aves em cativeiro reúne condições que favorecem a disseminação de doenças infecciosas, sendo a Salmonella uma dessas doenças infecciosas que acomete os psitacídeos. Portanto o objetivo do presente estudo foi isolar e identificar Salmonella spp. em Psittaciformes mantidos em criatórios comerciais e conservacionistas da Região Metropolitana de Fortaleza. Para o estudo, foram coletados swabs cloacais de 182 psitacídeos clinicamente sadios. Os resultados mostraram que três psitacídeos avaliados (1,65%) foram positivos: Amazona aestiva (Salmonella Lexington), Ara chloroptera (Salmonella Saintpaul) e Melopsittacus undulatus (Salmonella Newport). De acordo com a literatura científica, não há registro desses sorotipos em psitacídeos. Esta pesquisa evidenciou uma baixa prevalência de Salmonella spp. em Psittaciformes mantidos em criatórios comerciais e conservacionistas da Região Metropolitana de Fortaleza...

Activation of NFkappaB plays a pivotal role in many cellular processes such as inflammation, proliferation and apoptosis. In Drosophila, nuclear translocation of the NFkappaB-related transcription factor Dorsal is spatially regulated in order to subdivide the embryo into three primary dorsal-ventral (DV) domains: the ventral presumptive mesoderm, the lateral neuroectoderm and the dorsal ectoderm. Ventral activation of the Toll receptor induces degradation of the IkappaB-related inhibitor Cactus, liberating Dorsal for nuclear translocation. In addition, other pathways have been suggested to regulate Dorsal. Signaling through the maternal BMP member Decapentaplegic (Dpp) inhibits Dorsal translocation along a pathway parallel to and independent of Toll. In the present study, we show for the first time that the maternal JAK/STAT pathway also regulates embryonic DV patterning. Null alleles of loci coding for elements of the JAK/STAT pathway, hopscotch (hop), marelle (mrl) and zimp (zimp), modify zygotic expression along the DV axis. Genetic analysis suggests that the JAK kinase Hop, most similar to vertebrate JAK2, may modify signals downstream of Dpp. In addition, an activated form of Hop results in increased levels of Cactus and Dorsal proteins, modifying the Dorsal/Cactus ratio and consequently DV patterning. These results indicate that different maternal signals mediated by the Toll, BMP and JAK/STAT pathways may converge to regulate NFkappaB activity in Drosophila.

In tropical areas, local goats are often reported as being able to reproduce throughout the year, whereas an influence of season is found to be a factor when importing different dairy breeds. In these areas, oestrus synchronisation in goats is of interest for both technical (synchronisation of kidding, adjustment to forage availability or to continuous milk supply) and genetic reasons (dissemination of improved genotypes by AI). The use of a progestagen vaginal sponge combined with equine chorionic gonadotrophin (eCG)-cloprostenol injections remains an efficient tool to achieve synchronisation in temperate and tropical zones. However, the oestrus synchronisation treatments currently used for goats in tropical regions were originally developed for goats bred in temperate regions. For this reason, several alternative possibilities for improving the efficiency of the hormonal treatment are evaluated. Oestrus synchronisation with luteolytic agents is efficient (resulting in more than 70% of goats in oestrus) and it takes into account female cyclicity. In developing regions of the tropics, the use of buck teasing appears to be a promising approach to control oestrus and ovulation. The use of this technique provides 60% of females in oestrus within 5 days of introducing the bucks. Considering the availability of nutrients as the ultimate regulator of reproduction in the tropics, the control of nutritional condition is essential before the use of hormonal treatments for oestrus synchronisation in goats bred in these regions takes place.

BACKGROUND: The clonally derived rat prostatic epithelial cell line NRP-152 was examined to determine its ability to differentiate in a tissue recombination model. METHODS: NRP-152 cells alone, or combined with urogenital mesenchyme (UGM) or 10T1/2 fibroblasts, were grafted beneath the renal capsule of athymic rodent hosts. After 1 and 3 months, grafts were examined grossly and immunohistochemically. RESULTS: NRP-152 cells grafted alone formed small (10-25 mg) grafts without recognizable architecture. NRP-152 cells recombined with UGM formed larger grafts (50-100 mg after 28 days) containing glandular epithelium. Columnar luminal cells expressed cytokeratins 8 and 18 and rat prostatic secretory markers (DP-1 and DP-2). The epithelial ducts were surrounded by well-differentiated smooth muscle. The glandular epithelial cells were shown to be of rat origin. NRP-152 + 10T1/2 tissue recombinants formed small grafts (10-40 mg wet weight) after 1 month. The epithelial component of these grafts formed solid unbranched cords expressing cytokeratins 5 and 14; no glandular epithelial structures were observed. The stromal matrix was densely packed with a few cells expressing alpha-actin. CONCLUSIONS: A clonally derived prostatic epithelial cell line can form structurally and functionally normal prostatic tissue. This suggests that prostatic basal and luminal epithelial cells can be derived from a common progenitor.

OBJECTIVE: To assess the preference for sweetness among preschool children and differences between less and more deprived groups. In addition, to assess whether sweet taste preference was associated with presence of caries. METHODS: The sample was composed by 572 preschool children aged between 4 and 6, distributed in three day nurseries of varied socioeconomic background. Cross-sectional study developed in two steps. Preference for sweetness was assessed using a modified version of the Sweet Preference Inventory. The solutions varied in sugar concentration from 0 to 1,17 molar (0 to 400 g / litre). The presence of caries was assessed using the defs index. The socioeconomic status of the sample was classified according to the origin of domicile. RESULTS: The variation in preference for sweetness in our sample was too small. Most children preferred the sweetest juice. This reduced the ability of this variable to explain variation in caries prevalence. Despite this limitation, our results showed that socioeconomic level influenced preference for sweetness, which in turn was associated with caries prevalence. CONCLUSION: The socioeconomic status influence the sweetness preference and this, in turn, is associated with the dental caries prevalence.

Tonin- and kallikrein-like activities were investigated in different regions of the rat brain. The highest values of specific tonin activity, expressed as picomoles of angiotensin II liberated per minute per milligram of protein, were found in the neurohypophysis (359 +/- 190) and in the archicerebellum (200 +/- 68). The highest level of total tonin activity (picomoles of angiotensin II liberated per minute) was observed in the archicerebellum (902 +/- 308) which retained 97% of total tonin activity of whole cerebellum. Tonin activity was not detected in the cortex of cerebellum and in the choroid plexus. Low to intermediate values of specific (1.09 +/- 0.33 to 5.32 +/- 2.37) and total (1.38 +/- 0.55 to 93.00 +/- 49.30) tonin activity were observed in adenohypophysis, cerebellar nuclei, hypothalamus, thalamus, midbrain, pons, medulla and neurohypophysis. The lowest values of specific (0.11 +/- 0.05) and total (0.69 +/- 0.31) activities were observed in the hippocampus. Kallikrein-like activity was expressed as picomoles of p-nitroaniline liberated per minute per milligram of protein. No activity was detected in the neurohypophysis. For other regions, the values of the specific activity ranged between 72 +/- 18 and 282 +/- 14 except for the choroid plexus which was 5 +/- 2. The total kallikrein activity was also homogeneous ranging from 330 +/- 100 to 1870 +/- 112. For the choroid plexus and adenohypophysis the total kallikrein activity was 2.0 +/- 0.8 and 27 +/- 11, respectively.

Nineteen avaiable strains of Bradyrhizobium sp were used to test two forage legumes, Crotalaria juncea(Crotalaria) and Cajanus cajans (Guandu) for symbiotic N-fixation ability. Greenhouse experiments were conducted for nine weeks in order to evaluate shoot, root, and nodule dry weight, nodule number, and shoot nitrogen content. Nine strains presented relative nitrogen efficiency (RNE) over 100 per cent with Crotalaria cultivar, whereas only 3 strains were more efficient with Guandu. Strain 633 was the only which presented a high yield with both cultivars. Finally, plant dry weight accumulation followed N accumulation obtaining 0.6757 correlation coefficient for Crotalaria and Guandu, respectively

Oral health condition in samples of children from zero to six years old, examined in day nurseries for children from Bauru and S. Paulo County (Brazil) by the caries indicator in primary dentition-dmfs, is assessed. The first group did not receive oral health care where any as the second group received the standard oral care provided by the institution. Variables related to way of life and their relationship to the presence of caries were evaluated. Multiple regression analysis showed a statistically significant association of age and frequency of dental visits with the prevalence of dental caries (p < 0.05). The data also showed that 23.3% of the children from Bauru, and 9.3% of those from S. Paulo were free of caries, numbers that are very far from the 50% proposed by WHO for the year 2000. The overall prevalence of dental caries was greater for the Bauru group than for the S. Paulo group; however, statistical significance was only found in the 3-4 age-group (p < 0.05). Sex differences in the occurrence of dental caries were not statistically significant.

Functional differentiation of prostatic epithelium is manifested by the production of tissue specific secretory proteins. In vivo production of these proteins is dependent on the presence of serum androgens. A serum-free organ culture system was used to examine the initiation of prostatic epithelial cytodifferentiation in vitro using two rat prostate specific secretory proteins (DP-1 and probasin) as markers of epithelial cytodifferentiation. The dorsal-lateral and anterior prostatic (AP) lobes from 12-day-old rats were cultured for 6 days in serum-free medium in the presence or absence of androgens. At the start of culture, secretory proteins DP-1 and probasin were undetectable using Western blot analysis. DP-1 and probasin were produced by explants cultured in the presence of androgens but were not detected in the absence of androgens. Dose-response studies were carried out for testosterone (T), 5 alpha-dihydrotestosterone (DHT), 5 alpha-Androstan-3 alpha, 17 beta-diol (3 alpha-Adiol), and two synthetic androgens: 17 alpha-methyl-19-nortestosterone (MENT) and methyltrienolone (R1881). All androgens used were capable of inducing expression of DP-1 and probasin in vitro. T, R1881, and Ment were effective at doses of 10(-7) M to 10(-9) M, whereas both DHT and 3 alpha-Adiol were able to induce DP-1 and probasin at concentrations as low as 10(-10) M. Estrogen (17 beta-Estradiol), hydrocortisone (11 beta, 17 alpha, 21-trihydroxypregn-4-ene-3, 20-dione), and progesterone (4-pregnen-3, 20-Dione) were ineffective in inducing prostatic secretory activity. Hydroxyflutamide (alpha-alpha-alpha-trifluro-2-methyl-4'-nitro-m-lactoluidide ) blocked the induction of secretory activity elicited by T. From histological sections, it was observed that explants cultured with T exhibited tall columnar epithelial morphology with organized stromal components. Tissue sections of explants cultured without T exhibited a cuboidal to low columnar morphology with less organized stromal components when compared with glands cultured with T. A DNA synthetic index was established to measure proliferation in the explants at the end of the culture period. Explants cultured in the presence of T exhibited greater DNA synthetic activity than explants cultured in the absence of T (P < 0.05). Using this serum-free model, we can explore the mechanism for the initiation of secretory cytodifferentiation.

Bothrops jararaca venom was toxoided by stepwise iodination with cold iodine, and doses up to 30 LD50 were non-lethal by i.p. route (BICALHO et al., 1990). Groups of mice injected chronically with the native, or the iodinated venom, have been subjected to histological examination. In the native group, in the spleen, around the white pulp, an acellular, amorphous eosinophilic substance, metachromatic to Methyl Violet, PAS positive, and dichroic to Congo Red under polarized light, was present. Strong congestion in the liver, kidneys and lungs was found. The salivary glands were replenished with an amorphous substance in the serosal acini. The groups injected with the iodinated venom only show discrete alterations, more akin to the control group. The anavenin was immunogenic. Antibody generation in mice and rabbits was detected by ELISA. In mice, active protection against challenge with native venom was presented. The iodinated venom generated a rabbit antiserum with strong lines in gel immunoprecipitation against the lethal venom. A minimum neutralization titer of 2.3 mg ml-1 against the native venom was attained in the first cycle (28 days) of immunization. After 3 cycles (100 days), the protection rose to 5.1 mg ml-1.

RESUMO

This paper presents the results of a house-by-house survey to learn the occurrence of dental injury to the primary teeth in Brazilian children from ten to seventy-two months of age. The study sample comprised 576 children, 295 boys and 281 girls living on sixty-six streets selected in an urban area (Bauru) in the state of Sao Paul, Brazil. There were 174 children (30.2 percent) with positive reports of injuries to the primary teeth. The results show the need of an educational program in order to prevent the occurrence of dental trauma.

Sixty-eight clinical amalgam restorations were removed to examine the consistency of the calcium hydroxide bases. The variation of the base consistency was influenced by the age and/or clinical profile of teeth and the status of the restorations. The clinical conditions affected the calcium hydroxide base consistency.

RESUMO

The interproximal plaque-removing effect of three different materials (classic dental floss, rafia floss and indian floss) was tested in a group of 80 males (18 years-old). After two weeks all the materials had the same efficacy in removing the bacterial plaque, with significant reduction in interproximal plaque index. According to an opinion inquiry the rafia floss was the most comfortable material to use and has the lowest cost as well.

Thirty photoelastic dental-periodontal patterns were prepared according to the following criteria: ten with cilindrical shaped roots (series I), ten with conical shaped roots from apical do cervical (series II), ten with conical shaped roots from cervical to apical (series III), all of them having the same superficial area and apical basis. The photoelastic analysis showed that the main factor in determining the force distribution is the apical basis and that the conical roots from cervical to apical (series III) elicit a lateral compression on the periodontal support, which influences the magnitude of the force that is necessary to the formation of the some photoelastic deformations produced in the other series. The spectral distribution of the internal stress in the patterns of the series III showed the formations of incomplete fringes along with the lateral aspects of the roots. It was concluded that to make a comparison of the periodontal distribution of occlusal forces, it is of paramount importance the standardization of all steps concerning the construction of the patterns, the measurement of the photoelastic blocks and mainly the apical area of the root, which is the main determinant factor of the qualitative and quantitative results produced by the photoelastic analysis.
